It is true that the phrase separation of church and state does not actually appear anywhere in the constitution of the united states.

The law of love peace and liberty in the states extending to jews turks and egyptians as they are considered sons of adam which is the glory of the outward state of holland so love peace and liberty extending to all. Although the concept is older the exact phrase separation of church and state is derived from wall of separation between church and state a term coined by thomas jefferson. Though not explicitly stated in the first amendment the clause is often interpreted to mean that the constitution requires the separation of church and state.
The flushing remonstrance shows support for separation of church and state as early as the mid 17th century stating their opposition to religious persecution of any sort.
